Radiation-protective effects of piceatannol and acetylated resveratrol

Abstract: OBJECTIVE: To investigate radiation-protective effect of piceatannol and acetylated resveratrol. METHODS:CCK-8 Assay Kit was used to detect the survival rate of AHH-1 cells treated by 0,10,20,50,100 and 500 μmol/L piceatannol or acetylated resveratrol for 72 h,or by 4 Gy γ-ray for 48 h after treatment with 0,10,20 and 50 μmol/L piceatannol or acetylated resveratrol for 2 h. Kunming mice were treated with resveratrol,piceatannol and acetylated resveratrol before 60Co γ-irradiation. The average life span and the WBC count post-irradiation were observed. RESULTS:Compared to control cells (0 μmol/L),the survival rate of AHH-1 cells treated by 10,20 and 50 μmol/L piceatannol or acetylated resveratrol for 72 h was not changed (P>0.05),but the survival rate of post-irradiation AHH-1 cells treated by 10,20 and 50 μmol/L piceatannol or acetylated resveratrol increased (P<0.05 or P<0.01). After radiation for 24 h,the WBC count of middle-dose (100 mg/kg) and high-dose(200 mg/kg) acetylated resveratrol groups increased (P<0.05). Compared to control group (0 μmol/L),the average life span and the WBC count of radiation group decreased (P<0.05). Compared to radiation group,the average life span of mice in high-dose resveratrol or acetylated resveratrol groups increased. CONCLUSION:Piceatannol and acetylated resveratrol both had radiation-protective effect in post-radiation AHH-1 cells. In addition,acetylated resveratrol showed radiation-protective effect in post-radiation Kunming mice.
